Characterization of mouse salivary polypeptide secretion after oral administration of pilocarpine RChHN
LÓPEZ-SOLÍS,REMIGIO; PUENTE,MARGARITA; DURÁN,VIVIANA; MORALES-BOZO,IRENE; KEMMERLING,ULRIKE; PARDO,RODRIGO; WENK,CAROLINA.
Mouse salivary secretion provoked by single or multiple oral administrations of the cholinergic agent pilocarpine was characterized. An accessory ad hoc device, manipulated by a single operator, was used to collect saliva from various mice simultaneously. A single challenge by pilocarpine in the range 40-400 µg provoked a dose-dependent secretory response. The secretory response lasted for about 40 minutes and was constituted by four clearly defined stages, namely: lag (5-10 min), maximal flow (10 min), slowering (15-20 min) and postsecretory rest. In this response, usual parameters were: maximal flow rate, 30-40 µl/min; total volume of saliva, 250-350 µl/mouse and total salivary protein, about 700 µg/mouse. Temporal desensitization of the secretory...

Distribuição e caracterização anatômica dos coléteres foliares de Prestonia coalita (Vell.) Woodson (Apocynaceae) Rev. Bras. Bot.
RIO,MARIA C. SCATOLIN; CASTRO,MARÍLIA M.; KINOSHITA,LUIZA S..

Bras. Bot.
SILVA,ELAINE MARIA J.; MACHADO,SILVIA RODRIGUES.

Morphological changes in the vaginal epithelium during the oestrous cycle of Calomys callosus (Rodentia, Cricetidae) BJB
RODRIGUES,JEAN FÁBIO TORRES; FERRO,ELOISA AMÁLIA VIEIRA.
This study describes changes in the pattern of microridges, keratinization, desquamation, secretion, leukocyte infiltration as well as the increasing number of mitotic cells in the vaginal epithelium of Calomys callosus during the oestrous cycle. In proestrus, the epithelium is squamous and stratified with a fine layer of keratin and it is overlain by secretory prismatic cells. In oestrous, the epithelium is squamous, stratified and keratinized. In metoestrus, the epithelium is squamous and stratified with loss of the keratin layer. A leukocyte infiltration, extending from the base to the surface of the epithelial layer is also present. At the end of this phase, the surface cells start to become PAS-positive. In dioestrus, the epithelium is stratified. The...

Ocorrência e morfologia de glândulas nas pernas de Centris e Epicharis (Hymenoptera, Anthophoridae) Zoologia
Franco,Aldirene Costa; Cruz-Landim,Carminda da.
The presence of glands in insect legs is largely difunded. In bees they are present in solitary and social species. In Centris and Epicharis three different types of glandular structures were found: bag-shaped structures in males femur and tibia, unicelular glandular cells in ali segments of all leg pairs in males and females, but more developed in females, and differentiated epidermis in the basitarsus of both sexes. The tarsal gland present in the distal tarsomere of all legs of males and females are a special type of the bag-shaped structure. The nature of secretion and it use is unknow, but some morphological and stainning properties indicate a lipidic nature.

Production of recombinant proteins in Escherichia coli Genet. Mol. Biol.
Schumann,Wolfgang; Ferreira,Luis Carlos S..
Attempts to obtain a recombinant protein using prokaryotic expression systems can go from a rewarding and rather fast procedure to a frustrating time-consuming experience. In most cases production of heterologous proteins in Escherichia coli K12 strains has remained an empirical exercise in which different systems are tested without a careful insight into the various factors affecting adequate expression of the encoded protein. The present review will deal with E. coli as protein factory and will cover some of the aspects related to transcriptional and translational expression signals, factors affecting protein stability and solubility and targeting of proteins to different cell compartments. Based on the knowledge accumulated over the last decade, we...

Radioactive fucose as a tool for studying glycoprotein secretion BJMBR
Haddad,A..
The efficiency and reliability of radioactive fucose as a specific label for newly synthesized glycoproteins were investigated. Young adult male rabbits were injected intravitreally with [3H]-fucose, [3H]-galactose, [3H]-mannose, N-acetyl-[3H]-glucosamine or N-acetyl-[3H]-mannosamine, and killed 40 h after injection. In another series of experiments rabbits were injected with either [3H]-fucose or several tritiated amino acids and the specific activity of the vitreous proteins was determined. Vitreous samples were also processed by sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) and histological sections of retina, ciliary body and lens (the eye components around the vitreous body) were processed for radioautography. The specific...

Secretion of Streptomyces tendae antifungal protein 1 by Lactococcus lactis BJMBR
Freitas,D.A.; Leclerc,S.; Miyoshi,A.; Oliveira,S.C.; Sommer,P.S.M.; Rodrigues,L.; Correa Junior,A.; Gautier,M.; Langella,P.; Azevedo,V.A.; Le Loir,Y..
Lactococcus lactis, the model lactic acid bacterium, is a good candidate for heterologous protein production in both foodstuffs and the digestive tract. We attempted to produce Streptomyces tendae antifungal protein 1 (Afp1) in L. lactis with the objective of constructing a strain able to limit fungal growth. Since Afp1 activity requires disulfide bond (DSB) formation and since intracellular redox conditions are reportedly unfavorable for DSB formation in prokaryotes, Afp1 was produced as a secreted form. An inducible expression-secretion system was used to drive Afp1 secretion by L. lactis; Afp1 was fused or not with LEISSTCDA, a synthetic propeptide (LEISS) that has been described to be a secretion enhancer. Production of Afp1 alone was not achieved, but...

Secrets in secretions: genes that control nematode parasitism of plants Braz. J. Plant Physiol.
Hussey,Richard S.; Davis,Eric L.; Baum,Thomas J..
The most evolutionary advanced adaptations for plant parasitism by nematodes are the products of parasitism genes expressed in their esophageal gland cells and secreted through their stylet into host tissue to control the complex process of parasitism. Molecular analyses of nematode parasitism genes are revealing the complexity of the tools a nematode possesses that enable it to attack plants and paints a more elaborate picture of host cellular events under specific control by the parasite than previously hypothesized. Interestingly, the majority of the nematode parasitism genes discovered encodes proteins unique to plant parasites. Identifying the complete profile of parasitism genes expressed throughout the parasitic cycle of a nematode is the key to...

Testicular structure of three species of neotropical freshwater pimelodids (Pisces, Pimelodidae) Zoologia
Cruz,Rodrigo José Guimarães; Santos,José Enemir dos.
The testes structure of the following Siluriformes was studied: Pimelodus maculatus (Lacepède, 1803), Iheringichtys labrosus (Lütken, 1874) and Conorhynchos conirostris (Valenciennes, 1840). The testes of these species were dissected into cranial and caudal regions. The fringes of mature testes were counted and measured. Student's t-test was used to identify significant differences between fringe lengths of the two regions. To study the whole testes of each species in its resting and mature stage, testes were fixed in Bouin liquid for 6-8 hours and submitted to routine histological techniques. Standard histochemical techniques were used to detect carbohydrates and proteins. The testes of all species were paired and fringed organs. Histologically, cranial...
